High Performance Liquid Chromatography is a separation method based on the distribution of the components of a mixture between two immiscible phases, a stationary phase and a mobile phase. In liquid chromatography, the mobile phase is a liquid flowing through a column containing the stationary phase.

Mass spectrometric techniques that can be combined with liquid chromatography are Electrospray (ESI) or APCI (Atmospheric Pressure Chemical Ionisation). The coupled analysers are: triple quadrupole (low resolution) and QTOF (high resolution). In addition, some of the systems allow coupling with an on-line extraction system.
Types of analysis that can be performed:
- Qualitative: analysis of known or unknown compounds for the identification of compounds from a list of known compounds or from a search in a library of spectra. Statistical metabolomics studies can be performed on the basis of the data obtained.
- Quantitative: quantitative determination of one or more analytes of known structure in a sample, using Electrospray (ESI) or APCI as ionisation techniques.
